SCAR markers linked to Ms, a genetic male sterile gene in Chinese cabbage.

To identify molecular markers tightly linked to the male sterility gene Ms in Chinese cabbage,a BC1 population was developed from the cross between male sterile plants(MsMs)and fertile double haploid line 'FH-1'(msms).AFLP primers were screened for polymorphisms between the parents and two bulks representing fertile and sterile plants.Out of 512 primer combinations,3 combinations gave polymorphic amplification patterns.These polymorphic amplicons were cloned and successfully converted to dominant SCAR markers,designated syau_scr02,syau_scr03 and syau_scr05.All the SCAR markers are located on one side of the Ms locus with a map distance ranging from 1.60 to 2.50 cM.These SCAR markers show high polymorphism between 'AB01' and 12 fertile inbred lines.
