Cloning and expression of the porcine type I interferon receptor II extracellular domain gene preparation of polyclonal antibody

The cDNA sequence of porcine typeⅠ interferon receptor Ⅱextracellular domain gene was obtained from healthy piglets in pulmonary macrophage by RT-PCR. Then the cDNA gene was subcloned into the expression vector pET-32a. The recombinant plasmid was transformed into E. coli ( BL21/DE3) . The recombinant protein IFN2EC was highly expressed in E. coil( BL21/DE3) . BAL B / C mice were vaccinated IFN2EC recombinant protein and the polyclonal antibody of resistance for IFN2EC recombinant protein was acquired. The titer of polyclonal antibody was detected by ELISA and Western-blotting. The recombinant plasmid pET-IFNR2EC was successfully constructed in this study. The polyclonal antibody titer is 1: 12800 by ELISA detection. Western-blotting analysis showed the polyclonal antibody of IFNR2EC recombinant protein could combine with recombinant protein,which indicated IFN2EC recombinant protein had good immunogenicity.
