Immune response induced by DNA vaccine of metalloproteinases multi-antigenic epitopes

We aimed to research the influence of signal peptide on the immune responses induced by metalloproteinases multi-antigenic epitopes DNA vaccine in the eukaryotic expression system.The DNA sequences encoding metalloproteinases multi-antigenic epitopes were artificially synthesized.Then the sequences,with or without signal peptide linking,were linked to pIRESneo expression vector pIRESneo to construct two plamids for immuning mice.The result was detected by ELISA and the method of venom hemorrhagic activity neutralization.Both of two recombinant plasmids could induce antibody(1∶100) and relieve metalloproteinases-induced haemorrhage.Thus we can conclude that signal peptide dose not influence the metalloproteinases DNA vaccine expresession in mice.
