Cytotoxicity of quinocetone and olaquindox on human liver cells

To evaluate the cytotoxicity of quinocetone and olaquindox on human liver cells,Chang Liver cells and L-02 cells were treated with quinocetone and olaquindox.The cell growth inhibition rates were evaluated by MTT assay on the 4 h and 24 h,and the hormesis induced by low dose of the two drugs were detected too.On the 4 h,DNA damages were evaluated by single cell gel electrophoresis.All results were carried on the statistics and analysis.The inhibition ratio of quinocetone and olaquindox on Chang Liver cells were presented by 73.29% and 31.88%,and in L-02 cells were 76.51% and 39.7%,respectively.The result displayed dose and time dependent relationship;low dose of quinocetone can induce the hormesis on L-02 cells(P0.01);Quinocetone and olaquindox can significantly induce DNA damage on the two cells(P0.01),and olaquindox proved to be more deleterious than quinocetone.The results confirmed the cytotoxicity of quinocetone and olaquindox on human liver cells,and the toxic effects of quinocetone were smaller than that of the olaquindox.
