TRANSGENIC MAMMAL HAVING HUMAN Ig GENE LOCI INCLUDING A PLURALITY OF VH AND Vκ REGIONS AND ANTIBODY PRODUCED THEREFROM

2008
PROBLEM TO BE SOLVED: To provide transgenic animals producing humanized monoclonal antibodies and humanized monoclonal antibodies produced therefrom. SOLUTION: They are transgenic non-human animals operated so as to contain human immunoglobulin gene loci. In particular, animals in accordance with the invention possess human Ig gene loci that include a plurality of variable (VH and Vκ) genes regions. Advantageously, the inclusion of a plurality of variable region genes enhances the specificity and diversity of human antibodies produced by the animal. Further, the inclusion of such regions enhances and reconstitutes B-cell development to the animals, such that the animals possess abundant mature B-cells secreting extremely high affinity antibodies.
