Differentially expressed proteins and its significance of high and low metastatic human papillary ovarian cancer cell lines.

Objective To detect the distinct protein in two human papillary serous ovarian cancer cell lines with different metastasis potencies in vitro.Methods We respectively distilled total proteins of cell lysis solution and culture supernatant of two human ovarian cancer cell lines (HO-8910,a human ovarian cancer cell line,and its highly metastatic subline HO-8910PM.).Differentially expressed protein profile was analyzed by surface-enhanced laser desorption ionization time of flight mass spectrometry (SELDI-TOF-MS) on weak cation exchanger(WCX2) chips respectively.The protein profile was analyzed by Ciphergen proteinchip software 3.2.1.Results Eleven differentially expressed proteins were found in cell lysis solution between two cells,and of them,ten proteins was up-regulated and one protein down-regulated in HO-8910PM,comparing with that in HO-8910.Additionally,four over-expressed proteins were detected in the culture supernatant from HO-8910PM comparing with that from HO-8910.Interestingly,one differentially expressed protein,3144 at M/Z value,was found up-regulated in both the cell lysis solution and culture supernatant from HO-8910PM.Bioinformatic analysis from the SWISS protein database,we found that 3144 at M/Z protein profile may be one of signal transduction proteins CD24.Conclusion There are distinct proteins in protein profiling between high or low metastatic ovarian cancer cell lines.
