Effect of HSP70 on lung injury in rats with endotoxemia

Objective To explore the effect and possible mechanism of HSP70 on lung injury in rats with endotoxemia.Methods The rat model of endotoxemia was established with injecting lipopolysaccharide(LPS) through caudal vein.Twenty-four Wistar female rats were randomly divided into four groups:control group(C group),endotoxemia group(E group),Gln pretreated group(G1 group),Gln post-treated group(G2 group).All rats were killed at hour 6 after injecting LPS.Expression of HSP70,the activity of SOD and the contents of MDA in lung tissues were determined.The pathological changes of lung tissue were observed by light microscope.Results Compared with C group,HSP70 expression and the contents of MDA in the lung tissues significantly increased in E group(P0.01),the activity of SOD decreased(P0.01).Compared with E group,HSP70 expression and the activity of SOD significantly increased in G1 and G2 groups(P0.05),the contents of MDA decreased(P0.05),but there was no significant difference between G1 and G2 groups(P0.05).Conclusion The higher expression of HSP70 may protect lung injury in rats with endotoxemia,which may be related to the high HSP70-induced increase of anti-oxidation ability.But it could not be concluded that there is significant difference between Gln pretreatment and post-treatment.
