Cloning and sequence analysis of cytochrome b gene and SOD gene in Sitobium avenae under the ultraviolet induction.

【Objective】The purpose of this research is to confirm the results of early study and reveal the theory of Sitobium avenae's ecological geneties under the ultraviolet induction for 24 and 48h.【Method】According to the conserved regions of other known insects,apair of specific primers were designed and the RT-PCR method was used to amplify the partial sequences of cytochrome b gene and SOD gene fromS.avenae.The sequences were analyzed and the amino acid residues derived.【Result】Sequencing results showed that the cDNA fragment of cytochrome b gene was 420bp in size and it encoded 140amino acid residues,and the cDNA fragment of superoxide dismutas gene was 357bp in size and it encoded 119amino acid residues.The results displayed that the nucleotide sequence of cytochrome b gene in ones which were under ultraviolet induction 30W48h had three mutant sites different from contrast ones,and made 2 mutant sites on acid residues.And the nucleotide sequence of superoxide dismutas gene were the same between UV-induction ones and contrast ones.【Conclusion】Some specific mutant sites on cytochrome b gene of S.avenae under ultraviolet induction have been found.The mode of mutation is gene conversion or transversion.The SOD gene has no mutant sites.
