MRNA expression analysis of protein kinase D1 in cultured hypertrophic cadiocytes of neonatal rat affected by astragalus extract

【Objective】 To analyze mRNA expression of protein kinase D1(PKD1) in cultured hypertrophic cadiocytes of neonatal rat regulated by the astragalus extract and its mechanism.【Methods】 The cardiomyocytes of neonatal rat in primary culture were induced by Ang Ⅱ,and the model of hypertrophy cardiac myocyte was replicated.The mRNA expression of PKD1 in myocardial cells was analyzed from the perspective of different doses and different time,respectively.The former was randomly divided into normal control group,model group,valsartan(Val) in the control group,four different doses of astragalus extract groups.In the normal control group,any drug was added;in the model group,the final concentration of Ang Ⅱ was 10-6 g/L;in the Val group,the final concentration of Val was 10-6 g/L on the basis of the model group;the final concentration of different astragalus extract group were 10-4 g/L,5×10-4 g/L,10-3 g/L,2×10-3 g/L on the basis of the model group,respectively.The latter was randomly divided into normal control group,model group,Val in the control group,astragalus extract of 10-3 g/L dose group;the myocardial cells were collected at the time points of 24,48,72,96 h,and the mRNA expression of PKD1 was determinated by RT-PCR method.【Results】 Compared with the model group,the mRNA expression of PKD1 in the Val control group was significantly lower(P 0.05),as well as the astragalus extract of 5×10-4 g/L,10-3 g/L,2×10-3 g/L group in a dose-dependent trend.And the mRNA expression inhibition of PKD1 is roughly equal in the astragalus extract of 10-3 g/L,2×10-3 g/L dose group and the Val control group.While compared with the model group,the mRNA expression of PKD1 in the Val control group in different processing time(24,48,72,96 h) were significantly decreased(P 0.05);as well as the astragalus extract group in a time-dependent downward trend.And the mRNA expression inhibition of PKD1 is roughly equal in the astragalus extract of 72 or 96 h treatment group and the Val control group.【Conclusion】 Astragalus extract can significantly inhibit the mRNA expression of PKDl in hypertrophy cardiac myocytes induced by Ang Ⅱ,in a time and concentration dependence,which may provide a new research idea for the reverse of ventricular hypertrophy after myocardial infarction.
