The invention provides a culture medium of multipotent adult progenitor cells. The culture medium comprises 560 ml of low-sugar DMEM (Dulbecco Modified Eagle Medium), 400 ml of MCDB-201, 20 ml of fetal calf serum, 20 ml of double-antibody penicillin and streptomycin, 10 micro-grams of insulin-transferrin-selenium (1x), 0.039 mg of hexadecadrol, 25.6 mg of ascorbic acid-2-phosphate, 1 micro-gram of recombinant human epidermal growth factor, 1 micro-g of recombinant human platelet-derived growth factor BB and 1 micro-g of leukaemia inhibitory factor. A method for culturing multipotent adult progenitor cells by using the culture medium of multipotent adult progenitor cells disclosed by the invention comprises the following steps of: taking the culture medium of multipotent adult progenitor cells, performing low-density adherent culture of 500/cm , and then purifying the MAPC (Multipotent Adult Progenitor Cells) by removing CD45 and GlyA positive impurity cells by virtue of CD45 and GlyA immunomagnetic micro-beads. The culture medium of the multipotent adult progenitor cells provided by the invention is better for growing and higher in purity; the method for culturing the multipotent adult progenitor cells by using the culture medium of multipotent adult progenitor cells provided by the invention is shorter in culture time.
