RP-HPLC determination of trans-resveratrol and its related substances

Objective:To develop an RP-HPLC method to determine trans-resveratrol and its related sub- stances.Methods :The C_(18)column(5μm,150mm×4.6mm )was used as analytical column.Mobile phase con- sisted of methanol-0.1 mol· L~(-1)acetic acid(40:60),flow rate was 1.0 mL·min~(-1).The detection wavelength was set at 305 nm.Results:The standard curve was linear in the concentration range from 12 to 28μg·mL~(-1)for trans-resveratrol(r=0.9999).The limit of detection and quantitation for the assay were 0.09 and 0.29ng,re- spectively.Under the established conditions,the resolution between trans-resveratrol and its isomer was 7.20 and trans-resveratrol could also be well separated from its degradation products from acidic and basic hydrolysis and light exposure.Trans-resveratrol was sensitive to high temperature(60℃),light(4000 lx)and humidity (RH%:75%and 92.5%).Conclusion:The method is simple,accurate and sensitive,and can be used for the routine analysis of resveratrol.
