Immunological Identification of Sunflower ( Helianthus Annuus L.) Spherosomal Lipase

The spherosomal lipase from sunflower (Helianthus annuus L.) seedlings partially purified presented 2 protein bands with an approximate molecular weight of 61 and 66 kDa, in a characteristic zone of plant lipases. In order to determine which one corresponds to the lipase, we prepared specific antibodies against each protein bands. The polyclonal antibodies obtained have been tested by western blotting on spherosomal membrane proteins and also on a pure lipase from hog pancreas. The immunserum directed against the lower band (61 kDa) reacted specifically with the spherosomal lipase protein and is also reactive on hog pancreatic lipase. An enzymatic study showed that only the lower immunserum specifically inhibited sunflower lipolytic activity.
