Pathogenesis of atherosclerosis

Abstract The pathogenesis of atherosclerosis involves a complex interplay of endothelial dysfunction, lipid accumulation, inflammation, vascular smooth muscle cell proliferation, matrix turnover, and calcification. Genomic and epidemiological studies shed some light on the role of genetics in cardiovascular disease. It is appreciated that atherosclerotic lesions represent dynamic processes that evolve from fatty streaks to stable or unstable plaques. The clinical consequences of atherosclerosis, such as unstable angina, myocardial infarction or stroke, are a significant source of morbidity and mortality throughout the world. An understanding of the pathogenesis of atherosclerosis is important for understanding disease progression, the development of new therapeutics and ultimately the improvement of patient outcomes.
