Transcriptional regulation of T-bet, GATA3, ROR T, HERV-K env, Syncytin-1, microRNA-9, 192 and 205 induced by nisin in colorectal cancer cell lines (SW480, HCT116) and human peripheral blood mononuclear cell

Abstract Background Colorectal cancer is the third type of cancer, resulting in the fourth cancer mortality. Colorectal cancer can be related to loss of microbial balance. One of these bacterial species is Lactococcus lactis which can produce nisin. Methods In this study, SW480 and HCT116 cell lines and PBMC of healthy patients were treated with nisin to analyze T-bet, GATA3, RORγT, HERV-K env, HERV-W env, microRNA-9-5p, 192-5p and 205-5p expressions by quantitative real-time PCR. Results The expressions of microRNA-9-5p, -192-5p and -205-5p were elevated in SW480, HCT116 and PBMC. In PBMC, T-bet and RORγT had an increase. Also, there was an increase in HERV-W env expression in HCT116. Moreover, PBMC showed an increase in the expression of HERV-K env and a decrease in HERV-W env. Conclusions Nisin from Lactococcus lactis can change gene expressions, and inhibit colorectal cancer progression. Nisin should be added to diet to prevent the progression of colorectal cancer; however, further investigation is required.
