Direct observation of proton pumping by a eukaryotic P-type ATPase

P-type adenosine triphosphatases (ATPases) use the energy from ATP hydrolysis to pump cations across biological membranes. The electrochemical gradients that are generated control many essential cellular processes. Veshaguri et al. incorporated a plant proton pump into vesicles and monitored the dynamics of single pumps. Pumping was stochastically interrupted by long-lived inactive or leaky states. The work reveals how these proton pumps are regulated by a protein domain and by pH gradients. Science , this issue p.
