Process for releasing and isolation of n-glycan and o-glycan

1991
PURPOSE: To release N- and O-glycan from glycoconjugates. CONSTITUTION: The process for release of N- and O-glycan, and by adsorbing the released N- and O-glycans on chromatographic substance and eluting selectively, the process for isolating the glycan are provided. These processes are characterized by the following: glycoconjugates is placed under the influence of hydrazine agent (wherein the glycoconjugates are essentially salt-free and anhydrous, and the hydrazine is essentially anhydrous); the condition, where the glycoconjugates is placed under influence of hydrazine agent, N- and O- glycans are adsorbed by the chromatographic substance and are selectively eluted to release N- and O-glycan by a process which can recover the glycan in the essentially non-reducing and perfect form, is controlled.
