Pyridoxamine-A Natural AGEs/ALEs Inhibitor

2004
Aging and age related diseases, such as diabetes, atherosclerosis, and neurondegenerative diseases, are characterized mainly by increase in biochemical modification of tissue proteins. The Maillard reaction and advanced lipid peroxidation reactions are the most important reactions which cause the biochemical modification of tissue proteins, and then lead to formation of advanced glycation end products (AGEs) and advanced lipoxidation end products (ALEs) respectively.Based on the carbonyl stress hypothesis of aging,a variety of reactive carbonyl intermediates, the common products of the Maillard reactions and advanced lipid peroxidation reactions, can affect the oxidative modification of tissue proteins directly.Pyridoxamine,a naturally occurring B6 vitamin is recently found inhibiting the formation of AGEs/ALEs by trapping reactive carbonyl intermediates.Pyridoxamine is therefore promising to be a novel therapeutic drug for age related diseases.
