Determination of Urinary Cotinine in Annual Health Check-up Individual by Colorimetric Cotinine Kit

symptoms. Smoking, a risk factor of high blood pressure, dyslipidemia and cardiovascular diseases, are not included in an annual health check-up list. The aim of this study was to detect cotinine, a biomarker of smoking excreted in urine in males registered for an annual health check-up. The urinary cotinine assay kit was developed based on direct barbituric acid method. Color-intensity was visualized to determine smoking status comparing with that obtained by the immunological method. The urine samples used in this study were recruited from 110 males who underwent a medical examination at the Community Laboratory Services, Faculty of Associated Medical Sciences, Khon Kean University. The results showed 16 positive cases (14.5%). Twenty three samples were randomly taken to detect cotinine using the immuno-chromatography test strip. Sensitivity and specificity of the kit were 80% and 100%, respectively. Positive predictive value and negative predictive value were 100% and 94.7%, respectively. Positive agreement and negative agreement were 80% and 100%, respectively. The preliminary results show a high smoking status in male health check-up personnel. Urinary cotinine assayed by the test kit for smoking is accurate and reliable compared to the immunological method.
