Protection by ellagic acid, delivered specifically to the colon using microsphere capsules, of dextran sulfate sodium-induced colitis in rats

the effect of SPRA (D-Pro 4, D-Trp 7, 9) SP (4-11, Peptide Res.) on tumorous lesions in UC, SPRA in depot (5 mg/kg, decided by dose response study) was intramuscularly given to rats before and after DSS administration (2 weeks). DSS was given to rats for 3 months, Histological lesion score method (Onderdonk st al), myeloperoxidase (MPO) activity, colonic I L-1 (~ and -1/3 GRO/CINC-I(x and their mRNAs were determined. Results: Macroscopic and microscopic tissue injury, histological lesion score, the frequency of tumorous lesions, the elevation of MPO activity, IL-I~ and lp GRO/ CINC-I(~ levels were significantly suppressed by SPRA. Gone expression of IL-I,x and -1/3 GRO/CINC-I(~ were increased. SPRA inhibited development of tumors associated with UC, which was associated with the suppression of I L-1 (~ and -1/3 GRO/CINC-I(~ levels. Conclusion: The results suggest that the efficacy of SPRA in this model is attributable to the inhibitory action of the antagonist on the development of tumors with inflammatory mediators and leukocytes.
