The present invention discloses a method for preparing fluorescent probe and for detecting the pH of the special test kit. The present invention provides a structure for detecting the pH value of the fluorescent probe as shown in Formula Ⅰ. The present invention also provides a process for preparing compounds of the formula shown Ⅰ, comprising the steps of: shown dihydroxybenzaldehyde and morpholine by a reduction reaction (1) of formula IV, as shown in Formula Ⅴ substituted morpholin-methylphenyl diol ; (2) an inert atmosphere, in the presence of a base, as shown in formula ⅴ substituted morpholin-methylphenyl ⅵ glycol with the formula shown in tricarbocyanine dyes substituted decomposition rearrangement reaction, i.e., the formula ⅰ compound shown. The present invention provides a kit for detecting the pH value, comprising a solvent and a compound of the formula Ⅰ. Its specific fluorescent probe detection kit of the present invention provides a solution having a good response to pH, and enables determination of labeled lysosomes pH of lysosomes, a simple, low cost, rapid, sensitive, easy to advantage of the promotion and application.
