Objective To explore the method of immunohistochemiscal staining for HE slides after decoloration.Methods Dip HE slides in xylene to remove coverslip and glues;Rehydrate in alcohol gradient,rinse the slides in deionized water;Placed in 0.25% potassium permanganate for five minutes,rinse the slides in deionized water,bleached with 40.5% oxalic acid,rinse the slides in deionized water,dry the slides at room temperature;Drip one drop of PLLS on the tissue slides and bake to dry in oven at 60℃ about 60 minutes;Rinse slides three times in deionized water for 5 minutes;Use water-bath heating for antigen retrieval;Both control slides and tested slides use standard S-P method to have immunohistochemiscal staining.Results Positive staining were observed in both control slides and tested slides and all slides in good condition.Conclusion The method tested in this study is applicable for immunohistochemical staining in HE slides after decoloration.
