Genome editing in livestock

An efficient and easily reproducible in vitro culture system for porcine embryos meant for direct zygote microinjection was established. By injecting CRISPR/Cas 9 targeting the CMAH and GGTA 1 gene into IVF produced embryos successful gene editing was achieved. The usefulness of a novel dye lissamine green B for prediction of maturational competence of porcine oocytes was evaluated. To better characterise ovarian follicle, imaging of porcine ovarian follicles with single plane illumination microscopy was developed. Supravital dye brilliant cresyl blue was explored as contrast medium for imaging porcine ovarian follicle with multi spectral optoacoustic tomography.
