Effects of Sonication Assisted Agrobacterium-Mediated Transformation(SAAT) on Transient Expression of gus Gene in Explants of Soybean

We choose the variety of soybean"Dongnong 42"as the material in this experiment.The embryonic tip,cotyledonary-node and hypocotyls were treated with sonication under different power and processing time.The results showed that the optimal processing time and power for embryonic tip,is 3 min,80 W,and the efficiency of transient expreesion of gus gene is 87.5%.For hypocotyl or cotyledonary-node,the optimal processing time and power is 5 min,48 W,and the transient expression of gus gene is 75.0%and 66.7%respectively.In the comparison of treatment with sonication and cutting,the efficiency of transient expression of gus gene in explants treated with sonication are higher than those of treated by cutting.With sonication assisted Agrobacterium -mediated transformation,the efficiecny of transient expression of gus gene in embryonic tip,cotyledonary-node and hypocotyls were increased.
