Integrated Analysis of Proteome and Transcriptome Revealed the Landscape of Stroke Process Through MCAO Mice Model

2019
Stroke usually results in long-term disability and death worldwide. Recently, increased researches, both on physio-pathology mechanism and transcriptome, have highlighted the relationship between stroke process and immunity, especially inflammation. Here, we applied proteome study on MCAO mice model at 0h, 6h, 12h and 24h, by which a proteome profiling with 23 experiments were obtained and 41 differentially expressed genes were identified. Biology information study on our data discovered the importance of immune response, especially inflammatory response, cytokine-cytokine receptor interaction, innate immune response and ROS during stroke process. What's more, we also compared our data with multiple GEO datasets with or without time series, by which the similar pathways were identified and three proteins, including C3, Apoa4 and S100a9 were highlighted as a marker or drug targets for stroke, which were significantly up-regulated in MCAO model, both in our proteomic data and GEO database.
