Genetic diversity in three rainbow trout populations as revealed by SSR and RAPD

The genetic diversity of the populations for three cultivated populations of Oncorhynchus mykiss(50 from each population) is investigated by using the technology of random amplify polymorphic DNA(RAPD) and SSR.The result of RAPD show that 14 RAPD primers totally amplify 259 bands,the percentages of polymorphic loci(P) are 46.5%,33.3% and 47.8% respectively.Shannon index of phenotypic diversity H0 and Shannon index of diversity H are 0.734 7,0.094 2,0.623 9,0.079 9,0.763 8,0.097 9 respectively;And 51~105 alleles are detected from 18 SSR primer pairs.The number of effective alleles Ne is 1.03~6.50,the average observed heterozygosity H0 for the three populations are 0.603,0.601,0.430,and the average expected heterozygosity He are 0.665,0.547,0.498 respectively.It is concluded from the present study that there exists high genetic diversity in the three populations of Oncorhynchus mykiss.The differentiation of the populations is remarkable,Gansu rainbow trout is a special population of Oncorhynchus mykiss.
