Effect of Indirubin-3’-monoxime on cell proliferadion,cell cycle and Survivin expression in human esophageal cancer EC-1 cell line

Aim:To investigate the effect of Indirubin-3’-monoxime(IRO),a derivative of Indirubin and a potent inhibitor of cyclin-dependent kinases(cdk),on survivin expression in human esophageal cancer EC-1 cells,and to invest the antitumor mechanism of IRO.Methods:The EC-1 cells were treated with 1 μmol/L,5μmol/L and 10μmol/L of IRO. The proliferative status of cells was measured by methabenzthiazuron(MTT) assay. By fluorescence-activated cell sorting(FACS),the changes in the cell cycle were examined. The expression of survivin protein was determined by Western-blot.Results: IRO obviously inhibited growth of EC-1 cells in a dose-dependent and time-dependent manner(F=12.39,P0.05);Cell G0/G1 phase ratio decreased gradually(P0.05), S-phase ratio has not changed significantly, G2/M phase ratio increased gradually after treating with IRO(5 μmol/L) for 24,48 and 72 hours,cell cycle was blocked in G2/M stage in a time-dependent manner(P0.05); Survivin expression were down-regulated by IRO treating for 24 hours and down-regulated further at the 48 hours and 72 hours.Conclusion: IRO inhibits the proliferation of EC-1 human esophageal cancer cells via the cell cycle arrest in G2/M phase and survivin protein expression down-regulation.
