EFECTO DEL MEDIO DE FECUNDACIÓN IN VITRO SOBRE EL PATRON DE REACCION ACROSOMICA EN EL ESPERMATOZOIDE BOVINO

Frozen bull semen was thawed and diluted in three different in vitro fertilization media: TCM-199, TALP and BO. Assessment of cytoplasmicc membrane integrity and proportion of acrosome reacted sperm were accomplished by FITC-PNA lectin and PI staining in a epi-fluorescent microscope. The results showed that the number of intact, acrosome reacted and damaged membrane spermatozoa was significantly affected by both IVF medium and time of culture (p<0.001). So, the number of sperm with intact acrosome and membrane was higher when was cultured in TCM-199 (mean value 30.93+-1.18) than in TALP (28.79+-1.40) and BO (26.53+-1.31) and it was decreasing during the culture time. For the number of reacted acrosome spermatozoa was significantly higher for TALP than the others media and it was increasing during the culture period (mean value TALP 9.33+-0.81, TCM-199 6.76+-0.64, BO 7.33+-0.54),. In relation to died sperm, BO was significantly higher than the others media and the relation between the number of acrosome reacted and total intact membrane spermatozoa was lower for TCM-199 than the other; finally, the process of acrosome reaction was slower than BO and TALP.
