A computer assisted methodforthedetermination ofrestriction enzymerecdgnition sites

A computerprogramhas been developedwhichaidsin the determination ofrestriction enzymerecognition sequences.Thisisachievedby cleavingDNAs of known sequencewith a restriction endonucleaseand comparingthe fragmentation patternwith a computer-generated of patterns. The feasibility of this approach has been testedusing fragmentation patternsof0X174DNA producedby enzymesofbothknown and unknown specificity. Recognitionsequencesare predictedfortwo restriction endonucleases(BbvI and SfaNI) using thismethod. In addition, recognition sequencesare predictedfortwo othernew enzymes (PvuIandMstI)usinganothercomputer-assisted method.
