Eine neue Gefriertechnik für Präimplantations‐Embryorien

Inhalt: Fur die Tiefgefrierkonservierung von Mause- und Rinderembryonen wird eine einfache Gefrier- und Auftauapparatur beschrieben. Die Tiefgefrierung des Gefriergutes erfolgt in der Dampfphase eines offenen Stickstoffbehalters in einem variablen Abstand rum Flussigkeitsspiegel. Das Gefriergut ist lotrecht uber dem Behalter in einem fixen Drahtkorbchen deponiert. Eine vertikal bewegliche Hubvorrichtung hebt den Behalter, eine Styroporbox, an, wobei das Gefriergut in den Stickstoffdampf ver-senkt wird. Der Antrieb erfolgt iiber einen Elektromotor rnit Untersetzungsgetriebe und manueller Aussteuerung, womit Hub- und Gefriergeschwindigkeit reguliert werden. Aus den mit dieser Apparatur tieiqefrorenen Mause- und Rinderembryonen sind lebende Junge hervorgegangen. Contents: Freezing of mouse- and bovine embryos in nitrogen vapor of an open container above the liquid nitrogen level For the deep-freezing of mouse and bovine embryos an inexpensive freeze- and thaw apparatus has been constructed and used successfully. Controlled freezing of material has been achieved in the vapor phase of an open nitrogen container in variable distance to the liquid nitrogen level. The samples rest on a fixed small wire basket above the container. A vertical moveable lifting jack is lifting the container, a Styrofoam box, to burry the freezing goods in the vapor phase of the container. The speed of lifting and freezing is regulated manually by varying the revolutions of the small electric motor which drives the lifting jack via a reducing drive gear. After freezing and transfer of mouse and bovine embryos with this apparatus live offsprings have been born.
