Core-shell type upconversion luminescent labeling and graphene oxide luminescence resonance energy transfer based method for detection of ochratoxin A

2016
A core-shell type upconversion luminescent labeling and graphene oxide luminescence resonance energy transfer based method for detection of ochratoxin A is used for detection of ochratoxin A (OTA) in beer. The method is as below: coupling a core-shell type NaYF4:Yb0.18, Er0.02 @ NaYF4 upconversion luminescence nano material with avidin by an EDC / NHS reaction, modifying a biotinylated ochratoxin A aptamer to prepare a core-shell type energy donor probe; incubating the energy donor probe with OTA, wherein OTA aptamer preferentially binds to target OTA, then adding graphene oxide. The method uses the characteristics of overlapping of a graphene oxide absorption spectrum and a luminescence spectrum of core-shell upconversion luminescence nano material and DNA absorption; energy donor probe combined with OTA will not be adsorbed to the GO surface, and luminescence at 541nm is retained, and so as to realize the quantitative detection of OTA. The linear range is 0.001-250 ng/mL, and the detection limit is 0.001ng / mL. The invention used for OTA detection has the advantages of high sensitivity, fastness, simpleness and convenience, and can be applied in the detection of beer samples to obtain accurate and reliable results.
