Isolation and Functional Analysis of CesA Gene Promoter from Populus

The CesA gene promoter MDCesAP was amplified from triploid Chinese white poplar(Populus tomentosa Carr.).Its A/T composition is up to 63%.It has TATA box,polyA and so on regulatory elements in the sequences.The promoter showed lower similarity in the homologous sequences in comparison with the PTCesAP amplified from Populus tremuloids Michx.The MDCesAP was transformed into PCAMBIA1302 and PBI121 for substituting CaMV 35S promoter to study characterization of the promoter,the result indicated that the vectors were introduced into poplar,it was verified that MDCesAP is the xylem specific promoter.
