ISOLASI DAN KARAKTERISASI Bacillus sp. SEBAGAI KANDIDATPROBIOTIK DARI HUTAN MANGGROVE DESA MARGASARILAMPUNG TIMUR

Budidaya udang secara intensif telah memunculkan berbagai masalah seperti
penurunan kualitas perairan dan serangan berbagai penyakit oleh mikroorganisme
patogen yang berdampak pada kematian udang. Solusi dari permasalahan ini
adalah dengan menggunakan probiotik sebagai agen biokontrol. Salah satu
sumber diperolehnya bakteri kandidat probiotik adalah dari ekosistem hutan
mangrove. Tujuan dari penelitian ini adalah mendapatkan isolat Bacillus sp. dari
berbagai sampel biota mangrove tersebut dan karakteristiknya untuk digunakan
sebagai probiotik.
Berdasarkan hasil penelitian, diperoleh 5 isolat Bacillus sp. sebagai kandidat
probiotik yaitu KPP212, IP121, UJ131, UJ132, dan SB141 dari sampel biota
hutan mangrove Desa Margasari Lampung Timur. Karakteristik morfologi sel
isolat yaitu basil dengan sifat gram positif. Pada berbagai uji seleksi
menunjukkan seluruh isolat mampu menghidrolisis protein, tumbuh pada uji
cekaman pH (4-10), tumbuh pada uji cekaman salinitas dengan NaCl 0-6%, tidak
bersifat patogen. Pada pengujian biokimia seluruh isolat mampu memfermetasi
gula, non-motil, dan bersifat katalase negatif (kecuali isolat UJ132). Pada uji
kepekaan terhadap antibiotik isolat Bacillus sp. KPP212, UJ131, UJ132, SB141
sensitif terhadap kloramfenikol, asam nalidiksat, ampisilin, dan trimetoprim
namun resistan terhadap streptomisin. Sedangkan isolat Bacillus sp. IP121
sensitif terhadap trimetoprim dan resistan terhadap asam nalidiksat, ampisilin, dan
streptomisin.

abstract
An intensive shrimp farming has raised various issues such as declining in water
quality and invading of various diseases by pathogenic microorganisms causing
the death of shrimps. One of solutions to this problem is using probiotic as
biological control for intance, collected from mangrove ecosystem, one of which
is Lampung Mangrove Centre of Margasari Village East Lampung. The purpose
of this research is to obtain Bacillus sp. isolated from different samples of the
mangrove communities and to characterize them for probiotics use.
One way to obtain bacterial candidate for probiotics is selecting them. From this
research, 5 isolate Bacillus sp . are obtained and can be used as probiotics, namely
KPP212, IP121, UJ131, UJ132, and SB141. Characteristics morphology of
probiotic candidate are gram-positive cells and bacilli. From variety test
conducted for these probiotic candidate indicate that they are able to hydrolyze
proteins, grow at extreme pH (4-10) and at extreme salinity from 0-6% NaCl, and
have non-pathogenic activity. From biochemical testing indicates that probiotic
from isolates is capable of fermenting sugars, non-motile, and no catalase activity
(except isolate UJ132). In antibiotics sensitivity test, isolate Bacillus sp., such
as, KPP212, UJ131, UJ132, SB141 are sensitive to chloramphenicol, nalidixic
acid, ampicillin, and streptomycin but resistant to trimethoprim. While isolate
Bacillus sp. IP121 is sensitive to trimethoprim and resistant to nalidixic acid,
ampicillin, and streptomycin.
