Tissue culture and plant regeneration of Cinnamomum camphora Yongjin.

【Objective 】A micropropagation protocol was developed for Cinnamomum camphora Yongjin to provide technical support for large-scale multiplication of C. camphora Yongjin by tissue culture. 【Method 】Using auxiliary stems(1. 0- 1. 5 cm) of Cinnamomum camphora Yongjin as explants, adventitious bud induction, proliferation, rooting and so on were the focal points in the current research. 【Result】Bud inducement was initiated on the MS medium containing 1.0 mg/L 6-BA and 0.02 mg/L IBA. The inducement rate of valid buds reached 100%. The shoots were subcultured and multiplied on the improved MS medium containing 3.0 mg/L 6-BA and 0.05 mg/L IAA and the mean effective bud per shoot was 6.5. Harvested shoots were rooted in vitro in 1/2MS supplemented with 0.05 mg/L IBA and0.03 mg/L NAA for 30 days and the rooting rate was 87.6%.The survival rate was 90.0% when the rooting shoots were moved to the greenhouse. 【Conclusion】The combination of 6-BA(1.0 mg/L) and IBA(0.02 mg/L)is suitable to bud induction and growth for Cinnamomum camphora Yongjin, and the combination of 6-BA(3.0 mg/L)and IAA(0.05 mg/L)is beneficial to bud propagation. The combination of NAA(0.03 mg/L) and IBA(0.05 mg/L) is advantageous to rooting.
