Method for industrially producing recombinant human erythropoietin

2013
The invention discloses a method for industrially producing recombinant human erythropoietin. The method comprises multiplication culture and separation and purification. The method is characterized in that a serum-free medium is adopted for multiplication culture and is formed by adding 2-4% of Ultroser G, 250-300mg/l of sodium butyrate and 2-3mg/l of riboflavin into a dulbeccos modified eagles medium (DMEM). Two steps of chromatography are adopted for separation and purification, wherein the first step of chromatography is reversed phase chromatography, and the second step of chromatography is ion chromatography. According to the method for industrially producing recombinant human erythropoietin, the serum-free medium is adopted for the whole cell culture process, so that the defects of serum culture are avoided; and moreover, the production period is prolonged, the production quantity is increased, and the yield is improved. A double-step chromatography method is adopted, so that production links are reduced, the workload and cost are decreased, the yield is increased, and the total production recovery rate of the erythropoietin (EPO) can be more than 50%.
