Association of the interactions between IL-4 and Mina gene with children asthma

2011
Asthma is relative to multiple environmental factors and polygenetic backgrounds,so research on genetic background is significant to clarify the mechanism of asthma. We intended to study the associations of the polymorphisms of Mina rs4857304(G/T),IL-4 rs2243248(T/G) and rs2070874(T/C),and the interactions among IL-4 rs2243248,rs2070874,and Mina rs4857304 of children asthma in Chinese Han people.A case-control study was applied to observe SNPs distribution of IL-4 and Mina gene in the asthmatic patients and controls.202 cases of asthmatic children(aged 6.97±2.85 years) and 191 controls were included.SNP genotyping was performed by means of matrix assisted laser desorption ionisation-time of flight mass spectrometry method(MALDI-TOF).Multifactor dimensionality reduction(MDR) software was used to analyze the interactions among IL-4 rs2243248,rs2070874,and Mina rs4857304 of children asthma. The results showed that among the three loci, the Mina rs4857304 of asthmatic patients presented with a significantly higher portion of T allele frequencies(P = 0.0199,vs control;OR = 1.477,95%CI = 1.062-2.052),but no statistical differences among the genotypes frequencies of rs4857304 compared with controls(P = 0.0587).About IL-4 gene, the genotypes and allele frequencies of rs2243248 and rs2070874 have no statistical differences between the two groups. There were significant differences in the interaction model between IL-4 and Mina gene(including 2 and 3 loci best model) analyzed by MDR(P 0.05).In conclusion, our results highlight the role of Mina as a susceptibility gene for children asthma, and the interactions among IL-4 rs2243248 and rs2070874 and Mina rs4857304 also have associations with children asthma in Chinese Han people.
