Cloning of a DNA component of Banana bunchy top virus and analysis of its characteristic sequences

A new full nucleotide sequence of Banana bunchy top virus (BBTV) component 6 was cloned and analyzed. The sequence contained two main characteristic sequences, the CR-SL (Stem-loop common regions) and the CR-M (Major common regions). The phylogenetic tree constructed using the amino acid sequences of BBTV 6 suggested that BBTV contained two groups. The nucleotide and amino acid sequences of six BBTV components were aligned and compared, and the highest identities of the nucleotide and the amino acid sequences within each component were 96.10% and 98.27% respectively, while those among the six components were 70.78% and 56.24%, respectively. The phylogenetic trees based on full nucleotide sequences and amino acid sequences of the six BBTV components indicated six groups corresponding to each BBTV component. Based on CR-M nucleotide sequence, two groups corresponding to the places of origin could be divided. In addition, the secondary structures and some physicochemical properties of proteins of the six BBTV components were predicted. The result showed their significant differences.
