The therapeutic effectiveness of autologous bone marrow mesenchymal stem cells transplantation for acute renal failure in rabbits

Objective To evaluate the therapeutic effects of autologous bone marrow mesenchymal stem cells (MSCs) transplantation for acute renal failure in rabbits and the impact of the different transplantation opportunity on therapeutic effects. Methods Bone marrow MSCs were isolated and cultured in Vitro. Acute renal failure was induced by clamping bilateral renal artery for 90 rain,then the renal circulation restored. Thirty rabbits were divided randomly into three groups:transplanted group A (n = 10) ,transplanted group B(n = 10) and untreated group C (n = 10). In transplanted group A autologous MSCs labeled with BrdU were transplanted via carotid vein immediately after the resumption of circulation in the kidney,and in transplanted group B MSCs were transplanted 72 h after the resumption of circulation in the kidney. In untreated group C only the model of acute renal failure was made. Twenty-one days after the operation all rabbits were sacrificed. The survival rate among groups were analyzed, and renal function and renat structure were evaluated. Results Compared with untreated group C,MSCs transplanted groups have faster recovery of renal function and renal structure (P<0.05). The therapeutic effects of transplanted group A were better than those of transplanted group B (P<0.05). Conclusion Autologous bone marrow mesenchymal stem cells transplantation is effective for treatment of acute renal failure in rabbits. Bone marrow MSCs transplantation at early stage might have better therapeutic effects.
