Bioinformatics Analysis of Actin Related Protein 2/3 Complex Subunit 4 of Taenia asiatica

2007
Objective To analyze and predict the structure and characteristics of Actin related protein 2/3 complex subunit 4 (Arp2/3) of Taenia asiatica (T.a.ENO) by bioinformatics tools. Methods Using bioinformatics software package and tools of bioinformatics at webs sites of NCBI (http://www.ncbi.nlm.nih.gov/), ExPaSy (http://www.expasy.org/) were used to identify the genes and proteins of Taenia asiatica. The bioinformatics tools were also used to predict the physical-chemical characteristics, modification sites after translation, domains, subcelluar location, topological structure, second structures, and 3D structure of the enolase Taenia asiatica. Results A full-length cDNA sequence encoding the enolase of Taenia asiatica was identified. This novel cDNA sequence contains 718 bp with coding region from 30 to 530, encoding for 167 amino acids. The predicted amino acid sequence showed 78% identity and 90% similarity with that of Arp2/3 from Schistosoma japonicum. The protein, with a predicted molecular weight of 19533.9, contains no transmembrane region and subcellular location sequence. Three major linear epitopes located at 53~58, 74~82 and 138~143 were predicted. These three eiptopes are on the surface of spatial structure of Arp2/3. Conclusion Arp2/3 gene was screened from the cDNA library of adult Taenia asiatica by bioinformatical method.
