Application of reporter gene assay in functional study of hrnt-1 gene

Objective: To establish a reporter gene assay method for investigating the biological effects of hrnt_1 on the process of bronchial epithelial malignant transformation.Methods:To observe the localization of hrnt_1gene product in BEP2D cell in real time, the full_length cDNA was inserted into the pEGFP vector to express EGFP_hrnt_1 fusion protein. In addition,in order to observe the effects of hrnt_1 on key signal transduction pathways in vivo , the vectors containing cis_acting enhancer element and SEAP reporter gene were cotransfected with hrnt_1 expression vector.Results: The results showed that the protein of hrnt_1 located mainly in nucleus. Overexpression of hrnt_1 could activate the GR,c_My,EIK/STAT and NFκB signal transduction pathways.Conclusions:It is suggested that the reporter gene assay can be an effective method and adapt to the functional study of novel disease_related gene.
