Effect of Heat shock treatment on Embryogenesis for DH Production from Cauliflower Microspores

Stress treatment is a basic procedure to induce embryogenesis in microspore culture, especially in brassicas. In some cereal crops, Brassica napus, Brassica rapa and Brassica oleracea cold pretreatment also has been utilized. In the present study, the effect of heat shock treatment of 32°C for 48 hours followed by 25°C was investigated for direct embryogenesis in 10 Cauliflower (Brassica oleracea L. var. botrytis) cultivars. No heat shock treatment and shock treatment for more time were found to be detrimental on embryogenesis. The heat shock treatment of 32°C for 48 hours followed by incubation at 25°C significantly increased the rate of embryogenesis.
