Increased frequency of cancer in recent years and its impact on different physical, mental, and social dimensions of human life have turned it to a major problem of this century. We can group cancer according to the type of cells they start from viz. skin cancer, colon cancer, bladder cancer, breast cancer, prostate cancer, kidney cancer oral cancer etc. Among this, breast cancer is one of the most prevalent type of malignant neoplasm within women with more than one million new cases per year. In the recent years, with the help of targeted drug delivery system, several attempts were made to restrict the division, growth and proliferation of the mutating cancerous cells. As a matter of fact, small-molecular drugs are small enough to enter cells easily, so they are used for targeting inside of the cells. In this direction the use of Superparamagnetic Iron Oxide Nanoparticles (SPIONs) in breast cancer has a prominent role toimprovise the pharmacokinetic profile and demoting systemic toxicities of chemotherapeutic medicaments. Which ultimately results in selective targeting and delivery of the anticancer drugs to tumor tissues. In this review we discussed the targeting aspects of SPIONs for targeting breast cancer, pathophysiology of breast cancer, different mechanism of SPIONs for targeting and modification of SPIONs synthesis methods.
