The invention discloses a preparation method of a cartilage framework based on PVA (Polyvinyl Acetate) hydrogel, belonging to the field of biological tissue engineering. The method disclosed by the invention comprises the following steps of: firstly, establishing a CAD (Computer-Aided Design) mode of the cartilage framework, and printing a cartilage forming mould through a three-dimensional printer by taking hydroxyapatite microspheres as a base material according to the CAD mode; secondly, calculating mechanical properties of PVA hydrogel which differ in PVA percentage content by molecular dynamics, finding a satisficatory proportion, mixing uniformly the PVA hydrogel with an appropriate amount of osteoblasts under the guidance of the data to prepare PVA hydrogel containing the osteoblasts, and then filling the PVA hydrogel containing the osteoblasts in the mould; and finally, connecting the PVA hydrogel to an in-vitro dynamic perfusion system platform for culturing the PVA hydrogel, and taking the PVA hydrogel out of the mould for use after a cartilage framework is formed. Due to the method disclosed by the invention, the zero-pollution preparation of the framework is realized, the preparation period is shortened, and the mould has a certain gap structure so as to facilitate the circulation and the excretion of nutrient substances and metabolites of cells in the perfusion culture process of the framework.
