Mutation by using atmospheric and room temperature plasmas and high-throughput screening method for improving production of nosiheptide of Streptomyces actuosu

A novel atmospheric and room temperature plasma( ARTP) mutagenesis system was used by combining with the high throughput screening of 48-well microplates and inhibition zone method to obtain high-yielding mutants. The results showed that the correlation coefficients between the ratio of inhibition zone diameter and m A408,and m A408 between f A408 reached 0. 534 and 0. 896,respectively. Through several rounds of ARTP mutation and high-throughput screening,three genetic stable mutants were obtained with increase by 50% relative contents of nosiheptide than that of the control strain finally. ARTP mutagenesis techniques as an effective way to obtain high-yield nosiheptide strains was compared with the traditional shake flask screening method,48-well microplate and inhibition zone method could significantly improve the screening efficiency of high nosiheptide-producing strains.
