The expression of adiponectin and adiponectin receptor R2 mRNA in rats with nonalcoholic steatohepatitis and the intervention effects of telmisartan on it

2012
Objective To investigate the effects of telmisartan on the expression of adiponectin and adiponectin receptor R2 mRNA in rats with nonalcoholic steatohepatitis (NASH).Methods 35 male SD rats were randomly divided into normal control group(NC group,n=10),NASH model group(FC group,n=15) and telmisartan intervention group(FT group,n=10).The rats in NC group were given standard diet, however, the rats in the other two groups were given high fat diet for 16 weeks to induce NASH.After 12 weeks the rats in FT group were given telmisartan (5mg·kg-1·d-1) for 4 weeks by gavage. At the end of the 16th week, all the animals were sacrificed and the rat’s body weight was recorded, liver indexes were determined, the levels of ALT and AST in serum were detected by using automatic biochemistry analyzer, and the expressions of adiponectin and adiponectin receptor R2 mRNA in liver tissue were detected by RT-PCR.Results As compared with those in NC group, the levels of serum ALT and AST were significantly increased in FC group(P0.01),however,the expressions of adiponectin and adiponectin receptor R2 mRNA in liver tissue were significantly decreased in FC group (P0.01).As compared with those in FC group, the levels of serum ALT was obviously decreased in Ft group (P0.01),however,the levels of adiponectin and adiponectin receptor R2 mRNA in liver tissue were significantly higher than those in FT group (P0.05).Conclusion Telmisartan is effective in treating rats with NASH,and one of the action mechanisms is that telmisartan can promote the expression of adiponectin and adiponectin receptor R2 mRNA in liver.
