The Effect of Rhizoma Paridis on the Proliferation of Gastric Carcinoma Cell SGC-7901 Xenografts in Nude Mice

[Purpose] To investigate the effect of Rhizoma Paridis on the proliferation of human gastric cancer cell SGC-7901 xenografts in nude mice.[Methods] Human gastric carcinoma SGC-7901 cells were cultured and the nude mice xenografts models were established,and the nude mice xenografts models were randomly divided into control group,experimental groups with different concentration of Rhizoma Paridis(12.5,25 and 50mg/kg),chemotherapy group(5-Fu) and combination group(5-Fu+ Rhizoma Paridis).The changes of xenografts volume were measured.The expression of PDGF-B protein was detected by immunohistochemisty.[Results] Rhizoma Paridis could significantly inhibit the growth of gastric cancer xenografts in nude mice.The increase of xenografts volume in every treatment groups was less than that in control group(P0.05).The xenografts weight in every treatment groups was smaller than that in control group(P0.05),and the xenografts weight in other group was significantly larger than that in combination group(P0.05).These results suggested that Rhizoma Paridis combined with 5-Fu could significantly inhibit the growth of gastric cancer xenografts in nude mice.Compared to control group,the expression of PDGF-B protein was significantly decreased in every treatment groups(P0.01).[Conclusion] Rhizoma Paridis can inhibit the growth of human gastric carcinoma xenografts in nude mice,and there is a synergistic effect while Rhizoma Paridis combined with 5-Fu.The mechanism might relate to down-regulated expression of PDGF-B protein.
