Method for preparing aseptic plasma-protein containing fibrinogen and factor xiii

PURPOSE: To obtain the subject plasma-protein suitable as a component of a fibrin adhesive by subjecting human plasma to β-propiolactone treatment, ultraviolet irradiation, adsorption to anion exchanging material and then ethanol precipitation in two stages. CONSTITUTION: Plasma stabilized with a citric acid salt is treated with β- propiolactone, irradiated with ultraviolet rays and adsorbed to an anion exchange material to remove the factors II, VII, IX and X. The companion protein of the obtained solution is precipitated at -3 deg.C to a concentration of 9vol.% and the precipitate is separated by centrifugal separation and dissolved in a citrate buffer solution at pH6.35 and 37 deg.C to prepare a solution having a protein level of 13.3g/L. The companion protein is precipitated and removed with ethanol, a glycine citric acid buffer solution and a sodium citrate solution. Fibrinogen and factor XIII are precipitated from the residue by ethanol precipitation and dissolved in a citrate buffer solution at pH6.35 and 37 deg.C to a desired concentration.
