The study of the effects of resveratrol on TRAIL-induced apoptosis in human hepatoma cell line SMMC-7721

Objective The aim of our study was to observe the effect of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) on inducing apoptosis of human hepatoma SMMC-7721 cells,and resveratrol (Res) executing the role in the process.Methods SMMC-7721 cells were cultured in vitro.After being synchronized,the cells were divided into three gropus:control group,TRAIL intervention group,Res and TRAIL intervention group.The flow cytometry (FCM) was given to detect the effects of Res after the treatment with TRAIL/ Res and TRAIL for 24 hours.Results The apoptotic rate of SMMC-7721 cells of control group was 3.02% ±0.17%.The apoptotic rate was 8.55% ± 1.46%,with 100ng·ml-1 TRAIL.The apoptotic rates were 28.61% ± 1.41%,20.72% ± 1.33%,27.19% ± 1.43%,respectively,with the supplement of 25μmol· l-1 or 50μmol· l-1 or 100μmol· l-1 Res.The apoptotic index of SMMC-7721 cells in TRAIL intervention group was increased compared with control group(P < 0.01).Among res and TRAIL intervention group,the apoptotic index in 50μmol· l-1 and 100μmol· l-1 Res intervention group was increased compared with TRAIL intervention group(P < 0.01).But there no difference between 25μmol · l-1 Res intervention group and TRAIL intervention group (P > 0.05).Conclusions TRAIL can induce apoptosis of SMMC-7721 cells in vitro.Res can enhance TRAIL-induced apoptosis of SMMC-7721 cells in dose-dependent manner.
