The effect of murine serum amyloid A on tumor metastasis

Objective:To study the effect of all murine serum amyloid A(SAA) family members on tumor metastasis.Methods:We observed the expression of SAA1,2,3,4 mRNA in six tissues(brain,lung,liver,muscle,kidney and spleen)of LLC(Lewis lung carcinoma)-bearing mouse and normal mouse by real-time quantitative polymerase chain reaction.SAA1,2,3,4-GST fusion proteins were expressed and purified.Migration assay of LLC cells by SAA1,2,3,4-GST fusion proteins in vitro was used to evaluate the effect of SAA1,2,3,4-GST fusion proteins on migrated LLC cells.Results:The expression of SAA1,2,3,4 mRNA in six tissues(brain,lung,liver,muscle,kidney and spleen),especially in the lung,of LLC-bearing mouse were 0.15-47.29 times of that of normal mouse(P0.05)and SAA3 mRNA in the lung was highest among them.Murine SAA1,2,3,4-GST fusion proteins and GST protein were expressed and purified successfully.SAA1,2,3,4-GST significantly induced migration of LLC cells in vitro(P0.05).Conclusion:Murine SAA family might play a role in tumor metastasis and they could induce migration of LLC cells in vitro directly.
