상엽을 함유하는 한약재 복합물추출물(MAPC)이 세포 내 지방 축적 및 포도당 수송에 미치는 영향

This study was performed to evaluate effect of mixed aqua extract, consist of Mori Folium, Adenophorae Radix, Phllostachyos Folium, Citri Pericarpium (MAPC), on the lipid accumulation in 3T3-L1 adipocytes and glucose transport in L6 skeletal muscle cells. In inducing the differentiation of 3T3-L1 preadipocytes in the presence of an adipogenic cocktail, iso-butylmethylxanthine, dexamathasone, and insulin, we treated with MAPC. MTT assay for measurement of cell viability, oil red o staining for determination of lipid accumulation and western blot for analysis of Glut-4 (glucose transporter 4) protein expression were performed. There was no toxicity on 0, 0.05, 0.1, 0.5, 1 mg/ml of treated MAPC in 3T3-L1 and L6 cells. 0.2 mg/ ml of treated MAPC significantly reduced the lipid accumulation in 3T3-L1 cells compared with that of the same dose of single herbal extracts. And the Glut-4 expression was significantly increased in L6 cells treated with 0.2 mg/ml MAPC compared with that of control. These findings suggest that MAPC may be a potential therapeutic adjunct for obesity and diabetes by targeting the inhibition of adipogenesis in differentiated adipocytes, as well as the translocation of Glut-4 in skeletal muscle cells.
