Cultivo in vitro de bambu em diferentes sistemas de propagação

Dentre os novos sistemas de cultivo de plantas, os biorreatores apresentam destaque como um metodo de adequada aplicabilidade, o que pode proporcionar melhores condicoes para a multiplicacao de mudas em larga escala e com custo reduzido. O objetivo do presente estudo foi avaliar o numero de explantes estabelecidos (isentos de contaminacao bacteriana ou fungica), a multiplicacao e o alongamento de brotos de Bambusa vulgaris Schrad ex Wendl. em diferentes sistemas de cultivo. Explantes oriundos da fase de estabelecimento in vitro foram transferidos para tres sistemas de cultivo, sendo eles compostos por biorreator de imersao temporaria (BIT), cultivo in vitro em meio liquido(MEL) e cultivo in vitro em meio de cultura padrao (PADRAO), onde tambem foram testadas duas concentracoes de sacarose (0 e 30 g.L-1). Aos 14 dias, foi avaliada a taxa de explantes estabelecidos. Dos explantes considerados vivos e que nao apresentaram contaminacao foram avaliados o numero e o comprimento medio dos brotos. O maior numero de explantes estabelecidos foi observado no metodo de cultivo tipo BIT na ausencia de sacarose, e MEL quando suplementado 30 g.L-1 de sacarose. O BIT e MEL se destacaram apresentando maior numero medio de brotos por explante. O meio de cultura PADRAO resultou em menor numero de explantes estabelecidos e o menor numero de brotos por explante. In vitro BAMBOO CULTIVATION IN DIFFERENT SPREAD SYSTEMS ABSTRACT Among the new systems for cultivating plants, the bioreactors stood out as a method with suitable applicability, which may provide better conditions for the multiplication of seedlings in large scale and with a reduced cost. The aim of this study was to evaluate the number of established explants (free of bacterial or fungal contamination), as well as the multiplication and elongation of Bambusa vulgaris Schrad ex Wendl. sprouts in different cultivation systems. Explants derived from the in vitro establishment phase were transferred to three cultivation systems, which were composed by temporary immersion bioreactor (BIT), liquid-based in vitro cultivation (MEL) and in vitro cultivation in standard culture medium (STANDARD), where two sucrose concentrations were also tested (0 and 30 g.L-1). At 14 days, the rate of established explants was evaluated. From the explants considered alive and without signs of contamination, we evaluated the number and average length of sprouts. The greater number of established explants was observed in the BIT cultivation method in the absence of sucrose, and MEL when added 30 g.L-1 of sucrose. The BIT and MEL stood out showing greater average number of sprouts per explant. The STANDARD culture medium resulted in lower number of established explants, besides the lowest number of sprouts per explant . Keywords: Bambusa vulgaris , bioreactor, micropropagation, sucrose.
